| I. Background | |
| RARα（Retinoic Acid Receptor Alpha，視黃酸受體α）是核受體超家族成員，其結(jié)構(gòu)包 含高度保守的 DNA 結(jié)合域（DBD，含鋅指模體） 和 配體結(jié)合域（LBD，含配體依賴性轉(zhuǎn) 錄激活區(qū) AF-2），以異源二聚體形式（與 RXR 結(jié)合） 特異性識別靶基因啟動子上的視黃 酸反應(yīng)元件（RARE）；當(dāng)內(nèi)源性配體全反式視黃酸（ATRA） 結(jié)合 LBD 后，觸發(fā)構(gòu)象變 化，釋放共抑制復(fù)合物（如 N-CoR/SMRT），招募共激活因子（如 CBP/p300），進(jìn)而調(diào)控胚胎發(fā)育（神經(jīng)嵴分化、肢體形成）、細(xì)胞增殖/分化（如骨髓造血干細(xì)胞定向）及凋亡等 關(guān)鍵通路。 | |
